氮源對水稻田分離出念珠藻固氮酵素活性之影響

三種念珠藻(Anabaena sp.)已經由臺灣水稻田中分離出。CH1和CH3在實驗室內32°C,10 Klux,和空氣通氣培養下生長良好,細胞呈藍綠色。CH2於同樣條件下生長,則呈綠褐色。當培養基中硝酸塭,亞硝酸塭,或銨離子濃度超過0.1 mm時,三種念珠藻之固氮酵素活性均被顯著的抑制。當培養基中僅存有低濃度之亞硝酸塭,或銨離子(0.05 mM或0.1 mm)時,則會刺激CH1和CH2細胞內之固氮酵素活性,使其增高10-15%。

Three strains of Anabaena were isolated from Taiwan paddies. Strain CH1 and CH3 grew greenishunder 32°C, 10 klux, and air, but strain CH2 showed brownish color at the same growth conditions. Whenthe salt concentration of either nitrate, nitrite, or ammonium in the medium was over 0.1 mM, nitrogenaseactivity was significantly repressed in all three strains. Nitrite or ammonium at concentrations of 0.05 mMand .0.1 mM stimulated nitrogen fixation. It was suggested that this enhancement was due to not inactivat-ed nitrogenase activity in the cells after a short period of absorption and utilization of nitrite or ammonium.
