- 英文摘要: Glossogyne tenuifolia(hsiang-ju)(GT) is a traditionalantipyretic herb used in Chinese medicine; however, noinformation is available to explain its action. The objec-tive of this research was to elucidate the molecular phar-macological activity and the effective components in theethanol extract of GT. We found that GT had potent anti-inflammatory effects on the lipopolysaccharide(LPS)-activated murine macrophages, RAW264.7.GT down-regulated LPS-induced expression of inducible nitric ox-ide synthase(iNOS)by blocking its transcription.GT alsocaused a dose-dependent inhibition of the release ofprostaglandin E2 by repressing the promoter activity ofthe inducible cyclooxygenase(COX-2)gene.Moreover,GT exerted a dose-dependent inhibition of the LPS-stim-ulated release of the proinflammatory cytokines, TNF-α,IL-1(β, IL-6, and IL-12.To determine the mechanism bywhich GT inhibits LPS signaling,we focused on nuclearfactor-KB(NF-KB)activation. Western blot analysis re-vealed that GT abolished LPS-induced inhibitor-KB phos-phorylation. The electrophoretic mobility shift assaydemonstrated that GT abolished LPS-mediated KB DNAbinding activity. Moreover, macrophages were trans-fected with a vector coding for the luciferase reportergene under the control of NF-KB cis-acting elements, andthe transfected macrophages showed that the LPS-stim-ulated luciferase activity was GT-sensitive.These resultssuggest that GT attenuates inflammatory mediator syn-thesis of activated macrophages through an NF-KB-dependent pathway. The active components of GT wereidentified as oleanolic acid and luteolin-7-glucoside.Both of these compounds inhibited LPS-stimulated in-flammatory mediator production and NF-KB activation.We conclude that GT inhibits NF-KB-mediated gene ex-pression and downregulates inflammatory mediatorproduction in murine macrophages.
