甘遂一般藥理作用之研究

本研究觀察甘遂及炮製甘遂之乙醇提取液在不同動物之藥理作用,所得結果如下: 1.小白鼠腹腔注射藥物之一半致死劑量及95%可信限於甘遂為0.44公克/公斤(0.28至0.68公克∕公斤);於炮製甘遂為2.25公克/公斤(1.38至3.67公克/公斤)。 2.發現甘遂及炮製之甘遂均能抑制動物活動性測定儀及踏車法所測出之小白鼠運動活動性,而且發現對活動性測定儀所測得的運動活動性抑制現象較以踏車行為測得者顯著。 3.於麻醉貓:甲、甘遂及炮製甘遂對刺激坐骨神經所引起經骨肌及比目魚肌之收縮並無影響。乙、重複給予藥物可見動物有血壓升高,心律不整之現象,最後動物因而死亡。 4.於生理食鹽水導致利尿狀況之麻醉狗:甲、甘遂降低腎小球過濾率。乙、尿中鈉及氯離子濃度因服用甘遂而減少。丙、甘遂及炮製甘遂均使血壓、血液pH值下降。丁、炮製之甘遂使血中鈉與氯離子均增加,但未炮製之甘遂僅增加血中氯離子濃度。戊、動物給予甘遂或炮製甘遂均使實驗動物導致心律不整,繼之無尿,最後亦因之死亡。 5.大白鼠經胃管灌注藥物後,測定八小時內之排便量及大便含水量,發現未炮製甘遂使實驗動物排便量顯著減少,使大便含水量顯著增加。

The general pharmacological effects ofethanol extract of Kansui Radix (K. R.) andKansui Radix prepared (K. R. P.) were studiedin various species of animals. The followingresults were obtained: 1. Their median lethal dose (LD50) and95% confidence limited interval after intraperi-toneal injection into mice were 0.44g/kg (0.28-0.68g/kg) for K. R. and 2.25g/kg (1.38-3.67g/kg)for K. R. P. 2. Locomotor activity of the mice wereinhibited as determined by either ElectronicActivity Monitor (EAM) method or Rotarodmethod. It was found that both drugs couldinhibit the locomotor activity as determined byEAM method at lower doses than by Rotarodmethod. 3. In anesthetized cats: a) no effectswere found on the contractile force of Tibialisand Soleus muscles evoked by electricalstimulation of the sciatic nerve after administra-tion of K. R. and K. R. P.; b) elevation theblood pressure, cardiac arrhythmia and finallydeath were observed after repeated administra-tion of the drug. 4. In anesthetized dogs under salinediuresis: a) K. R. decreased the glomerularfiltration rate of the kidney; b) concentrationof sodium and chloride ions in urine weredecreased by K. R.; c) all the drugs decreasedthe pH value of the blood; d) concentrationof sodium and chloride ions in blood wereincreased by K. R. P., but K. R. increased theconcentration of chloride ion in blood only;e) cardiac arrhythmia, then anuria and finallydeath were observed after administration ofK. R. and K. R. P. 5. Total weight of stool and watercontent of the stool within 8 hours after drugadministration through a stomach tube in ratswere determined. It was found that K. R.decreased the excretion of stool, and alsoincreased the water content of the stool.
