- 英文摘要: In response to oxidative stress, the ischemic brain in-duces immediate early genes when its nuclear genescontain gene damage. Antioxidant that reduces genedamage also reduces cell death. To study the mecha-nism of neuronal sensitivity, we investigated the tran-scription of the c-fos gene after brain injury of the isch-emia-reperfusion type using focal cerebral ischemia-reperfusion in Long-Evans hooded rats. We observed asignificant (p<0.01) increase in c-fos mRNA in the isch-emic cortex immediately after brain injury. However, thec-fos transcript was sensitive to RNase A protectionassay (RPA) upon reperfusion. The transcript becamesignificantly resistant to RPA (42%, p<0.03) when 3-bro-mo-7-nitroindazole (25 mg/kg, i.g.), known to abolish nit-ric oxide, gene damage and neuronal sensitivity, wasinjected. Our data suggest that neuronal nitric oxide syn-thase and aberrant mRNA from genes with oxidativedamage cou{d be associated with neuronal sensitivity.
