以複乙二醇?沈澱劑之17β-雌二醇酵素免疫測定法

本報導?17β—雌二醇酵素免疫測定法之建立。以混合酸酐法將胡蘿蔔過氧化酵素與雌二醇-〔6〕-羧基甲基?共價結合,結合後平均?一酵素分子含1.7個雌二醇分子。酵素活性回收率?46% 。以雌二醇-〔6〕-羧基甲基?與牛血 清蛋白之複合物當做抗原引發雌二醇之兔子抗體血?。於首次抗原注射後約3個月抗體力價達到最高?。如此所得抗體很 具特異性,對於各種固醇類荷爾蒙之交叉反應試驗結果分別?雌酮1.89% ,雌三醇l.O % , 17α -雌二斷.08% ,睪丸酮 0.04%,助孕酮及雄酮小於0.01% 。酵素免疫測定法主耍利用複乙二醇當沈澱劑把與抗體結合之酵素-雌二醇複合物沈澱 出來。複乙二醇最終濃度?11% 。所得沈澱部分之酵素活性測定是以過氧化氫?基質,鄰二甲基對二氨基聯笨?氧之受質 。此法具有高靈敏度(70pg),同時具有快速及免除放射性危險等之優點。

An enzyme-labelled immunoassay for 17β-estradiol was developed. For this horse-radish peroxidase was conjugated with 17β-estradiol-6-carboxymethyloxime by mixed anhydride procedure. The number of 17β-estradiol residues bound per enzyme molecule was estimated to be 1.7 by radioimmunoassay, and the recovery of the enzyme activity was 46%. Antiserum for 17β-estradiol was raised in rabbits against 17β-estradiol-6-carboxymethyloxime: bovine serum albumin conjugate. The antibody titer reached maximum within approximately three months after primary immunization and was highly specific for 17β-estradiol. The cross reaction with estrone was 1.89%; estriol 1.0%; 17a-estradiol 0.08%; testosterone 0.04%; progesterone and androsterone <0.01%. During the assay, the separation of the antibody-bound from the free fraction of the enzyme label was performed by use of polyethylene glycol at final concentration of 11%. The enzyme activity of the bound fraction was determined with h2O2 as substrate and o-tolidine as oxygen acceptor. The assay possesses high sensitivity (70 pg) as well as the advantages of low cost and freedom from radiation hazard.
