- 英文摘要: The most frequently detected oncogene alterations, bothin animal and human cancers, are the mutations in theras oncogene family. These oncogenes are mutated oroverexpressed in many human tumors, with a high inci-dence in tumors of the pancreas, thyroid, colon, lung andcertain types of leukemia. Ras is a small guanine nucleo-tide binding protein that transduces biological informa-tion from the cell surface to cytoplasmic componentswithin cells. The signal is transduced to the cell nucleusthrough second messengers, and it ultimately inducescell division. Oncogenic forms of p21ras lead to unregu-lated, sustained signaling through downstream effec-tors. The ras family of oncogenes is involved in thedevelopment of both primary tumors and metastasesmaking it a good therapeutic target. Several therapeuticapproaches to cancer have been developed pointing toreducing the altered gene product or to eliminating itsbiological function: (1) gene therapy with ribozymes,which are able to break down specific RNA sequences,or with antisense oligonucleotides, (2) immunotherapythrough passive or active immunization protocols, and(3) inhibition of p21ras farnesylation either by inhibitionof farnesyl transferase or synthesis inhibition of farnesylmoieties.
