- 中文摘要: 以傳統目估法在顯微鏡下測定家畜動物的精子運動活力,由於其主觀性及準確度低,已不符實際上的需要。最近有一種原本為測定人類精子運動活力而設計的過膜泳動法已實質地克服了這些困難。本研究乃探討其在公豬精子運動活力測量上的應用性。本方法所使用的裝置很簡單,它只由注射針筒、過濾膜及改造的細胞培養皿所組成。精液放入該裝置於37℃下培育三小時後,測定游過濾膜精子的比例。以本方法測定之精子運動活力和目估法測得之活力有很高的相關性。使用8μm大小的濾膜所得測量值較使用5μm濾膜者為可靠。本方法適用於精子濃度在1.5×10 /毫升至6.0×10 /毫升的精液樣品之活力測定。本研究所得結果顯示此過膜泳動法為一簡便,客觀,可量化而且具再現性的測量公豬精子運動活力的方法。
- 英文摘要: The conventional microscopic methods for evaluating sperm motility of domestic animals are mostlyinadequate due to their subjectivity and lack of precision. Recently, a trans-membrane migration method,originally developed for the examination of human sperm motility, has substantially overcome theseproblems. This study investigated the applicability of the method to boar sperm motility measurement.The apparatus used was simple and consisted only of syringe plungers, poriferous membranes, and modifiedmulti-well culture plates. It measured the proportion of sperm in the semen that moved across the mem-brane after incubation at 37℃ for 3 hr. The sperm motility as measured by this method correlated wellwith that measured by direct microscopic examinations. The measurement was more reliable using an8-μm instead of a 5-μm pore-size membrane. The method was found to work eauallv well for the spermmotuity measurement or the semen witn a sperm concentration between 1.5 × 10 /ml and 6.0 × 10 /ml.The results indicate that this method is a simple, objective, quantitative, and reproducible design for themeasurement of boar sperm motility.
