動控制,構成一連續流動式測定系統;於常溫下,當液體流速為7.0 cm/min時,進行一次樣品之測定僅須五分鐘,並具
極佳之重現性。此酵素電極對麩胺酸溶液(pH 4.5)之線性應答濃度範圍為0.1-1.0 g/L,其操作安定性約為700次連續分
析;於4°C下貯存45天,電極之應答特性仍相當穩定。
本文同時比較〝單層膜模式〞與〝雙層膜模式〞之理論應答,以解析酵素膜與 (方程式無法摘錄) 透氣膜之擴散阻力對電極應答特性
之影響。結果為:〝雙層膜模式〞所預測之理論應答與酵素電極之實際應答相當?合,並確認酵素膜與 (方程式無法摘錄) 透氣膜之擴
散阻力對電極之應答同具重要性。
t - 英文摘要: A cross-linking L-glutamate decarboxylase electrode has been developed for a continuous flowmeasuring system. A microcomputer was applied for automatic control and calibration of the system. Theelectrode demonstrated very good linearity in the region for 0.1-1.0 g/L L-glutamic acid and reached itsmaximum response potential within 5 minutes at 7.0 cm/min fluid velocity. The electrode showed goodstability for at least 700 continuous assays of pure L-glutamic acid solution and 250 assays of dilutedfermentation broth. It also had excellent reproducibility and stability after 45 days storage. A double layer model has been proposed to study the effect of diffusion resistances of (方程式無法摘錄) withinboth the enzyme membrane and gas permeable membrane. The results obtained in this study indicate thatthe diffusion resistance of (方程式無法摘錄) in the gas permeable membrane is much larger than that in the enzymelayer and cannot be neglected.
