- 英文摘要: The heterotrimeric GTP-binding regulatory proteins (Gproteins) play an important role in the regulation ofmembrane signal transduction. Recently, we identifiedthe association of Go protein with mitotic spindles. Herewe have investigated the relationship between Go pro-tein and microtubules. We used temperature-dependentreversible assembly and taxol methods to purify micro-tubules from bovine brains. Goa and Gβ proteins wereidentified in the microtubular fraction by both methods. The Goα subunit in the microtubular fraction could beADP ribosylated by pertussis toxin. Co-immunoprecipi-tation data also revealed that Go protein can interactwith microtubules. Exogenous Go protein could be in-corporated into the assembled microtubular fraction,and 5 g/ml (60 nM) of Go protein inhibited 40% ofmicrotubule assembly. Western blot analysis of Goα-1and Goα-2 in microtubular fractions showed that onlyGoα-1 is associated with microtubules. We conclude thatthe Goα-1β proteins are associated with microtubulesand may play some role in regulating the assembly anddisassembly of microtubules.
