- 英文摘要: Microarray technology was used to gain an insight intothe molecular events of tumor cell growth inhibitionmediated by the soy isoflavone genistein. For this, a sus-ceptible bladder tumor line TCCSUP was treated with theinhibitory dose (50 M) of genistein for various periodsof time, followed by mRNA isolations, cDNA probe prep-arations, and hybridization individually to cDNA chipscontaining 884 sequence-verified known human genes.After analyzing the hybridization signals with a simplequantitative method developed by this study, we de-tected that egr-1, whose expression has been associatedwith proliferation and differentiation, was transiently in- duced and this expression pattern was later confirmedby RT-PCR. Thus, microarraytechnology is a reliable andpowerful tool for profiling gene expression patterns inmany biological systems related to cancer. We furtherdetected many groups of genes with distinct expressionprofiles and most of them encode for proteins that regu-late the signal transduction or the cell cycle pathways.These genes warrant further investigation as regardstheir roles in the susceptibility of the tumor cell line to theantitumor drug.
