- 英文摘要: Tissue factor (TF) is an important regulator and effectormolecule of coagulation in various inflammatory states.In sepsis, expression of TF by activated endothelial cellsleads to disseminated intravascular coagulation.Scopa-rone is extracted from the traditional Chinese herb Ar-temisia scoparia and is known to have potent anti-inflammatory properties. In the current studies, we ex-amined the effects of scoparone on inhibiting lipopoly-saccharide(LPS)-induced TF expression in cultured hu-man umbilical vein endothelial cells(HUVECs).Flow-cytometric analysis revealed LPS(10 pg/ml)-activatedsurface TF induction was concentration-dependently in-hibited by scoparone(10-400 pM.The concentrations ofscoparone used in this study did not affect cell viability.The elevation of the procoagulant activity of TF by LPSwas suppressed by scoparone.The LPS-induced super-oxide formation was markedly decreased by scoparone.Messenger RNA expression of TF in LPS-activatedHUVECs was also reduced by scoparone. Furthermore,scoparone did not significantly affect the IkBa degrada-tion.Our results demonstrate that the inhibition of sco-parone on LPS-induced TF expression in HUVECs maymediate by the mechanisms suppressing superoxide an-ion formation and TF transcription.
