水稻花葯培養之研究 花粉時期與低溫處理

日本型栽培稻臺南五號及H-124的花粉依其發育分為減數分裂、早期單核、中期單核、晚期單核、第一次有絲分裂及雙核六個時期。將含上述時期花粉的花葯置改良式White培養基上,在溫度25±1℃及光期十六小時(1000 1ux)的環境下培養,發現中期單核花粉最易形成癒合組織,產生癒合組識的花葯的頻度臺南五號為39.2%,H-124為38.0%。含早期及晚期單核花粉的花約亦可產生癒合組識,但頻度甚低,尤以臺南五號為然。臺南五號含單核花粉的花葯經低溫處理後,產生癒合組織的花葯頻度較未處理者高,但是這種差異在各處理中沒有明顯的趨勢。因為每個處理所用的花葯數目太少,而且同一處理中花葯內花粉的時期亦可能有某些程度的差異,所似低溫處理所得到的結果被認為是由取樣造成的,在統計上可能不具意義。

Anthers of two varieties of rice,Tainan 5 and H-124, were cultured ona modified White's medium at differentstages of pollen development to deter-mine the optimal stage. for callus for-mation. The results showed that onlyuninucleate pollen grains were capableof developing into calluses, whereasmicrosporocytes and binucleate pollenfailed to grow. When the uninucleatestage was further divided into three sub-stages, the early-, mid-, and late-uni-nucleate stages, the peak response occur-red at the mid-uninucleate stage; beforeor after this stage the response declinedsharply. Immature inflorescences of Tainan5 were to exposed to temperatures of5℃ and 10℃ for 24 and 48 hours. An-thers containing uninucleate pollen werethen cultured on a modified White'smedium to determine whether low tem-peratures may have an effect on thesubsequent growth of the pollen. It wasobserved that anthers treated with lowtemperatures produced calluses withslightly higher frequencies than the con-trol. However, the results cannot be in-terpreted to mean that low-temperaturetreatment has a positive effect on callusformation. The reasons for this are dis-cussed.
