人體胎盤甲硫胺酸活化?對動力學機轉

The order of substrate binding and product release of human placental methionyl-tRNA synthetasewas investigated in the aminoacylation reaction by the initial velocity and product inhibition experiments.The kinetic patterns obtained from the present studies and those from our previous studies in the ATP-PP exchange reaction are consistent with a Bi(random) Uni Uni Bi (ordered) ping-pong mechanism, with dead-end complex formation both by PP and AMP, the first and last product, respectively. The proposed kineticmechanism is similar to that for human placental threonyl-tRNA synthetase (Pan, et al., Int. J. Peptideprotein Res., 20, 159, 1982) rather than that for rat liver methionyl-tRNA synthetase (Pan, et al., J. Chin.Biochem. Soc., 4, 17, 1975).
