顯示ALDH2失活型之國人在飲酒後,胃之乙醛氧化速率會較活性型者為低。A - 英文摘要: Aleohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) isozyme phenotypes weredetermined in surgieal and endoseopie biopsies of the stomaeh and duodenum by agarose isoeleetriefocusing. γ-ADH was found to be the predominant form in the mueosal layer whereas β-ADH was predo-minant in the muscular laver. Low-K ALDH1 and ALDH2 were found in the stomach and duodenum.High-K ALDH3 isozymes occurred only in the stomach but in the duodenum. The isozyme patterns of gastric mucosal ALDH2 and ALDH3 remained unchanged in the fundus, corpus, and antrum.The stomaeh ALDH3 isozymes exhibited a value for acetaldehyed of 75 mM, and an optimum for acetaldehyed oxidation at pH8.5. Since the value was high, ALDH3 contributed very little, if any, to gastrie ethanol metabolism. The activities of ALDH in the gastrie mueosa deficient in ALDH2 were60-70% of that of the ALDH2-active phenotypes. These results indicate that Chinese lacking ALDH2activity may have a lower acetaldehyde oxidation rate in the stomach during alcohol consumption.
