- 中文摘要: 本論文以測量細胞核體積的大小來探討長期持續光照對雌鼠下視丘神經細胞形態之影響。本研究共使用12隻年齡為 16 至 18 個月之 Long-Evans 雌鼠為實驗材料。6隻動物為實驗組,另6隻則作為對照組。實驗組動物給予持續光照刺激長達 11 個月之久,而對照組動物則置放在照明週期(光照 14 小時,黑暗 10 小時)規則的動物房內。動物的食物及飲水皆不加以限制。動物斷頭犧牲之後,迅速取下腦組織,以Bouin液固定,製成石蠟包埋之切片,以甲酚紫(Cresyl Violet)染色,在光學顯微鏡下測量下視丘四個區域神經細胞核的直徑。再根據之公式,計算細胞核之體積V,A為細胞核之最大直徑,B為垂直於A之直徑。本實驗所量測的下視丘部位,包括下列四個區域:視叉上核(Suprachiasmatic Nucleus),室旁核(Paraventricular Nucleus),腹內核(Ventromedial Nucleus),和視前區(Preoptic Area)。統計分析測量所得結果,發現除視前區外,實驗組的其餘三個下視丘區域神經細胞的核體積皆顯著小於對照組。 雌鼠在長期持續光照下,會引起體內兩種激素-松果腺素(Melatonin)及女性素(Estrogen)含量明顯的變化,其與下視丘神經細胞形態改變的關係,在本報告中作了詳細的討論。
- 英文摘要: A light microscopic quantitative analysis was performed for studying the effects of long-term expo-sure to continuous illumination on hypothalamic morphology in female rats. Nuclear volumes of neuronsin the suprachiasmatic nucleus, paraventricular nucleus, ventromedial nucleus and preoptic area weremeasured in 16-18 month-old Long-Evans female rats which had been exposed to continuous light for 11months. Significant nuclear volume shrinkage could be observed in all investigated regions except for thepreoptic area in these animals when compared to that of control rats kept under a photoperiod of 14hlight and 10h dark. The relationship between morphological changes of hypothalamic neurons and themarked alteration of melatonin and estrogen levels induced by long-term exposure to continuous light isdiscussed.
