- 中文摘要: 酵素顆粒內之擴散阻力通常相當嚴重而不能忽略,擴散與反應同時存在也使得酵素反應外顯之動力學表現異於其真實行為,在酵素光學分割系統中亦然,當質量傳送之影響不能忽略時,酵素外顯之鏡像選擇性與其真實之鏡像選擇性往往有很大之差距。針對粒內擴散不容忽視之酵素光學分割系統,本文提出改良式暫態回應法用以求取酵素之真實鏡像選擇性。理論分析之結果顯示,在酵素過量之情形下,酵素之真實鏡像選擇性可逕由被分割基質之殘餘量及系統之 Thielem modulus求得。雖然所得之關係式與Lopez等人所提出之關係式相同,但由於本文所提之方法適用範圍更廣、操作簡單、且數據之處理分析容易,應是探討酵素真實鏡像選擇性之利器。
- 英文摘要: The internal diffusion resistance in enzyme articles is often significant and cannot be neglected. The coexistence of diffusion and reaction makes the apparent kinetic behavior of the enzyme- catalyzing reaction deviate from its intrinsic characteristics. Also in a kinetic resolution system in which the effect of mass transport cannot be ignored, the apparent enantioselectivity might be very different from the intrinsic enantioselectivity. The present study proposes a modified transient method which was shown to be able to determine the intrinsic enantiomeric ratio of a biocatalytic process when the influence of internal diffusion is considered. According to the theoretical prediction, the enzyme's intrinsic enantiomeric ratio can be calculated directly from the amount of the resolved substrates eluted and the Thiele modulus of the system, provided that an excess amount of the enzyme is employed to force the system following a linear kinetics. Although the obtained relation is the same as that of Lopez et al. (1990), the present method provides higher applicability. The method presented herein can expedite the evaluation of the intrinsic enantiomeric ratio, which is related to the potential of enzyme for achieving kinetic resolution, owing to its ease of operation, simplicity of data analysis.
