- 英文摘要: Heat shock protein(HSP) is a family of highly conservedproteins that respond to stress and participate actively incytoprotection.Within the HSP family, HSP70 is the ma-jor inducible member that confers protection against celldeath.This study investigated whether HSP70 plays aneuroprotective role at the rostral ventrolateral medulla(RVLM),the origin of sympathetic neurogenic vasomotortone in the medulla oblongata where the organophos-phate insecticide mevinphos(Mev) acts to elicit cardio-vascular toxicity. Experiments were carried out in adultmale Sprague-Dawley rats that were maintained underpropofol anesthesia.Intravenous administration of Mev(960 g/kg)induced a significant increase in the HSP70level in the ventrolateral medulla during phase I(‘pro-life' phase), and returned to baseline during phase I(‘pro-death’phase)Mev intoxication.Compared to artifi-cial cerebrospinal fluid,normal mouse serum(1:20),orsense hsp70 oligonucleotide(50 pmol)pretreatment, mi-croinjection of an anti-HSP70 antiserum(1:20) or an anti-sense hsp70 oligonucleotide(50 pmol)bilaterally intothe RVLM significantly increased mortality, shortenedthe duration of phase I intoxication and augmented theinduced hypotension in rats that received Mev (960 g/kg,i.v.).These results suggest that HSP70 induced in theRVLM during Mev intoxication provides neuroprotectionagainst the organophosphate poison via prevention ofcardiovascular depression.
