- 中文摘要: Streptomyces violaceoruber produces, a pigment-antibiotic that inhibits Gram-positive bacteria but neither Gram-negative bacteria nor true fungi. For this reasonit can not be used presently for practical purpose. In order to employed the principleof recombination for Streptomyces violaceoruber, the present paper will report theresults of intraspecific recombination.1. Polyauxotrophic mutants of Streptomyces violaceoruber 3 (Histidine , Methio- nine ) and 2 (Proline , Cystein ) were isolated after ultra violet irradiation of the conidia of the wild-type strain. The mutation rate was 1/5.3x .2. Mating conidia of 2 with that of 3 in complete medium, after 7 days, replicate in minimal medium, then 56 "primary parental isolates" were obtained.3. Inoculated conidia of "primary parental isolates" colonies in complete medium until conidia have developed, replicated in minimal medium which supplemented with different four amino acids in combination (Proiine: P, Cystein: C, Histidine: H, Methionine: M), the recombinants for different amino acid medium were obtained as following: C+H: 31.8%, C+M: 16.9%, C+P: 78.6%, M+H: 9.4%, P+M: 79.6%, P+H: 74.1%,C+H+M: 23.8%, C+H+P: 88%, C+P+M: 82%, M+H+P: 90%, C+H+M+P 96.5%, minimal: 12.4%.4. Inoculated they parental type conidia of "secondary parental isolates colonies" in complete medium until conidia have developed, then processed the same manner as regular procedure, the recombinants for different amino acid medium were as following: C+H: 14.3%, C+M: 7.8%, C+P: 39.8%, M+H: 45.4%, P+M: 32.6%, P+H: 47.9%, C+H+M: 44.3%,C+P+H: 54.7% C+P+M: 51.1%, M+H+P: 55.2%, C+H+M+P: 60.9%, minimal medium 17.4%.5. The compatibility system in Streptomyces violaceoruber may be characterized as follows. When conidia of each of two freshly isolated polyauxotrophic mutants are mated on complete medium, a small number, usually 10 to 100, of "primary heterokaryotic colonies" appear. On the other hand, when" secondary parental isolates" (derived from the conidia produced by primary heterokaryons) are mated, the frequency of heterokaryon formation is increased by a factor of ons several times. This high-frequency mating is reasonably stable property, only after a considerable number of subcultures. The results from this study, it is concluded that Streptomyces violaceoruber exhibit intraspecific genetic recombina- tion and posses the phenomenon of heterokaryosis.
