- 英文摘要: The cell polarity gene, crumbs (crb), has been shown toparticipate in the development and degeneration of theDrosophila retina.Mutations in CRB1, the human homo-logue of Drosophila crb, also result in retinitis pigmento-sa and Leber congential amaurosis. In this study, weused the gain-of-function approach to delineate the rolesof crb in developing Drosophila eye.In the third-instarlarval stage, eye development is initiated with photore-ceptor differentiation and positioning of photoreceptornuclei in the apical cellular compartment of retinal epi-thelium.In the pupal stage, differentiated photorecep-tors begin to form the photosensitive structures, therhabdomeres, at their apical surface.Using GMR-Gal4todrive overexpression of the Crb protein at the third-instareye disc, we found that differentiation of photoreceptorswas disrupted and the nuclei of differentiated photore-ceptors failed to occupy the apical compartment. Usinghs-Gal4 to drive Crb overexpression in pupal eyes re-suited in interference with extension of the adherensjunctions and construction of the rhabdomeres, andthese defects were stage-dependent. This gain-of-func-tion study has enabled us to delineate the roles of Crb atselective stages of eye development in Drosophila.
