- 中文摘要: 本研究在於訂定老鼠內池A粒子(IAP)基因81二端LTR部份的DNA鹼基序列,將所得之結果與一些已發表的IAPLTR之DNA鹼基序列詳細比對後發現本IAP 81之LTR構造和一般IAP LTR之構造以及一些因子之特性都很類似,但是諸LTR之DNA鹼基序列則有較明顯不同之處。在IAP 81 LTR中亦發現了各種典型的特性,包括有代表著LTR構造內之各種轉錄信號如Promotion, Initiation, Polyadenylation 和 Termination, 以及在整個DNA鹼基序列中較為特殊之處如 Z-DNA 區,Enhancer core sequence 和 Glucocorticoid-responsive element. 對於一LTR之一級構造可能之生化意義及重要性,本篇報告亦作了更進一步的討論。
- 英文摘要: The intracisternal A-particle (IAP) genome is a retrovirus-like gene family. Over 1000 copies ofIAP genes are dispersed throughout the mouse genome. The purpose of this study was to determine thenucleotide sequence of both ends of the long terminal repeat (LTR) units which blnd the IAP gene 81.The results obtained showed that. whereas the structural domain and several genomic characteristics of theIAP 81 LTR units and other LTR structures of a given IAP gene were almost identical, the nucleotidesequences of these LTR units associtiated different IAP gene were sometimes very different fromeach other. In the comparisons of different LTR structures. the transcriptional signals for promotion,initiation, polyadenylation and termination as well as several characteristic sequences of the Z-DNA region, enhancer core sequence and glucocorticoid-responsive element were all found to be present in IAP 81LTRs. The possible roles of the primary nucleotide sequence of an LTR structure are discussed.
