本試驗在探討幾丁質水解菌之分佈、特性及其對蝦殼水解能力。在所試試樣中發現,蝦體和土壤所含之幾丁質水解菌
菌數為CFU/g,約佔總生菌數的2.7%,其中不具蛋白?(包括酪蛋白?及膠原蛋白?)之幾丁質水解菌,僅佔
幾丁質水解菌總數的7.4%,且均分佈在土壤中,在蝦體並未被發現。不具蛋白?之幾丁質水解菌,在nutrient broth、try-
ptic soy broth等液體培養基無法生長。在本試驗所開發使用之salts-pectin broth液體培養基中,則所分離之幾丁質水解菌
均能生長,但缺乏或MgSO及果膠時,所分離之幾丁質水解菌均無法生長,這些幾丁質水解菌均為異營菌,其
最適生長pH為8.0,大部分菌株在pH 7.0以下即無法生長。若干具有蛋白?及幾丁質?的分離菌株對蝦殼的水解能力比單
獨具有幾丁質?、單獨具有蛋白?或混和單獨具有幾丁質?及單獨具有蛋白?兩株菌者為強(p<0.05);其中有一株
Aeromonas caviae(D1株)最強。
e - 英文摘要: This investigation was conducted to understand the distribution, characteristics and the hydrolyticactivity of chitinolytic bacteria on shrimp shells. The counts of the chitinolytic bacteria in soil and wholeshrimp were enumerated. The counts ranged from to CFU/g, which comprised 2.7% of the totalbacterial count. Among them, 7.4% of the chitinolytic bacteria could not produce caseinase and gelatinase.The chitinolytic bacteria that could not produce caseinase and gelatinase were found in soil but not inshrimp, and could not grow in nutrient broth or tryptic soy broth. A salts-pectin broth was developed forcultivating chitinase producing bacteria. All of the isolates could grow in this medium, except when or pectin were removed from it. The optimun pH for growth was 8.0 for the isolated chitino-lytic bacteria, but some strains could not grow when pH was below 7.0. Some bacteria which could produceboth chitinase and protease had higher shrimp-shell liquefaction activity than did those which could produceonly chitinase or protease, or a mixture of both strains (p<0.05); among them, Aeromonas caviae (strainD1)was the highest.
