- 英文摘要: Previously, we developed a TraT display system to ex-press snake venom rhodostomin(RHO), a disintegrin, onthe external surface of Escherichia coli [J Biomed Sci6:64-70;1999]. To show anew potential use of the TraTdisplay system, we employed a biotin labeling techniquecoupled with SDS-PAGE and flow cytometry analyses tofurther demonstrate and confirm the expression of TraT-RHO on the E. coli surface. We also showed that theexpression of TraT-RHO on the cell surface not only facil-itated the bacteria adhesion to BHK-21 cells but alsoinduced bacterial internalization into BHK-21 cells. Thisfeature allowed us to enrich the TraT-RHO expressionbacteria about 10,000-fold starting with a mixture ofTraT-RHO bacteria with β-galactosidase-positive bacte-ria in a ratio of 10 :10 through four cycles of BHK-21 cellendocytosis and replating of engulfed bacteria on agarplates. We therefore suggest that the TraT display sys-tem can be applied to select out bacteria expressing aspecific peptide sequence from a large population of dis-play library through the process of receptor-mediatedendocytosis and reamplification cycles.
