反應之檢定中,此抗體僅與胰島前素有17.5%之反應,此為意料中之理想結果。在「結合圖線」分析時,發現此抗體之
結合能力甚均勻,其Kd為75 × M,結合容量則為10× M。所標定之追?劑,其「特異活性」約為150μ
Ci/μg,優於購自國外廠商者。在4℃與人血清培養三天,追?劑之完整性並未受損。本測定怯之敏感度為2.5μU/ml。
用人胰島素或豬胰島素做追?劑,與人或豬胰島素做各種組合之標準曲線時,發現四種組合之標準曲線均大致平行。其
中似乎以人胰島素追?劑與豬胰島素配合所得之標準曲線最為敏感,且斜率最大。
訊 - 英文摘要: Pure porcine insulin was used to immunize guinea pigs, and antisera with high titer (~ 1:400,000)and excellent sensitivity and specificity were obtained. Semi-synthetic human insulin and porcine insulinhad been used either as a tracer or a standard to characterize these antisera. The sensitivity of the assay wasaround 2.5 μU/ml. The antisera had no cross-reactivity with guinea pig insulin, TRH, Somatostatin, LHRHand Secretin. The cross-reactivity with c-peptide, glucagon and porcine proinsulin were 0.025%, 0.14% and17.5%, respectively. A Scatchard plot revealed a straight line with a Kd of 7.5 × M. The serum dilu-tion and recovery studies showed good parallelism with the standard curve. The averaged recovery of serumimmunoreactive insulin (IRI) was 101 ± 3.96%. The intra-assay and inter-assay C.V. were 7% and 9%,respectively. An incubation study showed no damage of the tracer in serum at 40℃ for up to 72 hours. Thecomparisonal studies using either human or porcine tracer in combination with either human or porcinestandard revealed no difference of the standard curves. The human tracer seemed better than the com-mercial porcine tracer in terms of sensitivity and specific activity. The results of using human tracer witheither human or porcine standard showed a good correlation in measurement of serum IRI. A GlucoseTolerance Test (GTT) showed normal response of serum IRI in a normal subject, and virtually no responsein a patient with insulin dependent diabetes mellitus.
