內生菌根孢子接種對紅檜幼苗生長之影響

本試驗研究之目的,在以六種內生菌抱子接種至紅檜幼苗,視其與紅檜苗形成內生菌根之能力及對紅檜幼苗生長之影響。 將紅檜種子以30%H O 殺菌25分鐘後,以無菌蒸餾水洗淨,隨即播種至無菌之玻璃管媒體內生長至五個月時,即分別移植至殺菌過之泥炭土—蛭石—水苔混合營養媒體及含磷酸三鈣之砂土中。每一移植孔接種50個孢子,對照者加孢子之濾液10 c.c.。本試驗採用完全逢機設計,每一處理重複十次。苗木培養於光度為5,000 Lux,光週期16小時,日溫23℃,夜溫17℃之生長控制室中。苗木每隔七天加營養液。經接種三個月後,每隔一個月作菌根感染之確定。將根以自來水洗淨後,切成1~2 cm長之片斷,旋放入90℃ 10% KOH溶液中50分鐘,經弱鹼性 H 0 漂白及酸化後,以乳酸酚藍染色。由本試驗可得下列兩點結論: (1)A. laevis, A. trappei, G. macrocarpus與G. microcarpus之內生孢子,皆可與紅檜幼苗形成內生之瘤囊菌根。 (2)紅檜幼苗能與G. macrocarpus較快形成內生菌根,且感染率較其它處理者高,因之其對高生長,地上部乾重、根乾重及促進氮、磷、鐵之吸收,有顯著〈p<0.05)之效益。

The objective of this study was inoculateTaiwan Red Cypress seedlings with six differentendomycorrhizal species, to check their abilityof endomycorrhizal formation and their effectson the growth of these seedlings. The seeds of Taiwan Red Cypress collectedfrom Ta-Shie-Shang were sterilized in 30%H 0 for 25 minutes, than they were washedthree times with distilled water thoroughly.These seeds were sown in sterilized mediumcontained in sterilized medium contained insterilized glass tube. They were transplantedinto sterilized peat: vermiculite: moss nutrientmixture medium and sterilized sand contained500 mg Ca (PO ) respectively, until the seedlingswere five months old. Each transplanting holewas added with 50 spores; and for the control,it was added with 10 cc of spore filtrate filteredfrom 50 spore solution. The completely randomized design was usedin this experiment, and each treatment wasreplicated 10 times. These seedlings werecultured in a growth chamber under the con-ditions of light intensity 5,000 Lux, photoperiod16 hours, day and night temperatures: 23℃ and17℃ respectively. Seedlings were watered withmodified Melin-Norkran's nutrient solution every7 days. After three months of inoculation, themycorrhizal infection was examined, everymonth. All roots were washed with tap water,then cut into 1~2 cm section and set in 90℃10% KOH solution for 50 minutes. After thesections were bleached and acidified, they werestained with lactophenol blue and washed withlactophenol. From this study, two conclusions wereobtained as follows:(1) All the endospores of A. laevis, A. trappei, G. macrocarpus and G. microcarpus could from vesicular mycorrhizae with Taiwan Red Cypress seedlings.(2) The Taiwan Red Cypress seedlings could form endomycorrhizae with G. macrocarpus much faster, and their infected percentage was much higher than that of any other treatment. Therefore, its effects on height growth, top dry weight, root dry weight, and on the nutrient uptake of nitrogen, phosphorus and iron were significant (P<0.05).
