- 英文摘要: Neuronal transplantation has provided a promising ap-proach for treating neurodegenerative diseases. Recent-ly, efforts have been directed at in vitro induction of var-ious stem cells to transform into neurons. We report thefirst successful quantities in an in vitro attempt at direct-ing the transformation into neurons of human umbilicalmesenchymal cells, which are capable of rapid prolifera-tion in vitro and are easily available. When cultured inneuronal conditioned medium, human umbilical mesen-chymal cells started to express neuron-specific proteinssuch as NeuN and neurofilament(NF)on the 3rd day andexhibited retraction of the cell body, elaboration of pro-cesses, clustering of cells and expression of functionalmRNA responsible for the synthesis of subunits of thekainate receptor and glutamate decarboxylase on the 6thday. Between the 9th and 12th days, the percentage ofhuman umbilical mesenchymal cells expressing NF wasas high as 87%,while functionality was demonstrated byglutamate invoking an inward current. At this stage, cellswere differentiated into mature neurons in the postmito-sis phase.
