- 中文摘要: 許多研究顯示,Retinoids對於上皮細胞癌具有延緩及抑制生長之作用,然而對於腦瘤之作用則尚未確立。另一方面,具維生素A活性之Carotenoids也具有抗癌細胞之活性,但不具維生素A活性之Carotenoids是否有延緩及抑制癌細胞生長之作用則尚未被研究。因此,我們比較4種Retinoids(Ral, RA, RAc, RP)及3種Carotenoids(包括具維生素A活性之BCT,及不具維生素A活性之LCP及CCT)對老鼠C-6 glioma細胞(一種以N-nitrosomethyl urea誘生腦瘤衍生而來的細胞株)之抑制作用。 結果顯示所有的Retinoids及Carotenoids在1.0 μg∕ml之濃度時均會抑制C-6 glioma細胞之生長,Ral及RA有較強的抑制作用(抑制70-80%),其他Retinoids或Carotenoids則有50-60%的抑制作用。若以plating efficiency來估計活存50%時之濃度,RA及RAc為0.1 μg/ml,其他依序為Ral, LCP, RP, BCT及CCT。其抑制作用係由於這些化合物對細胞DNA的合成抑制作用,但對RNA及蛋白則較少影響。 綜合以上結果顯示,Retinoids及Carotenoids對老鼠腦瘤細胞有延緩及抑制之作用,其作用以Retinoids較強,而不具維生素A活性之Carotenoids亦有抑制作用,顯示Carotenoids抑制癌細胞之生長,不完全是因為轉換成維生素A之作用,而有其他可能之機轉。
- 英文摘要: The inhibitory effects of 4 retinoids, namely, retinal (Ral), retinoic acid (RA), retinyl acetate(RAc),and retinyl palmitate (RP), and 3 carotenoids including beta-carotene (BCT), lycopene (LCP), and crocetin(CCT) on the growth and DNA synthesis of rat C-6 glioma cells were studied. All the retinoidsand carotenoids caused reduction of plating efficiency and inhibition of the cellular growth. RA was themost potent inhibitor of plating efficiency, followed in decreasing order by RAc, Ral, LCP, RP, BCT,and CCT. The effects of various doses of retinoids and carotenoids on the inhibition of DNA synthesiswere clearly demonstrated in the growing C-6 glioma cells, whereas negligable effects of these compoundson the RNA and protein synthesis were observed. These results suggested that retinoids or carotenoidsare biologically active as anti-tumor agents against brain tumor cells in culture, while carotenoids appearedto be less active.
