利用掃描電子顯微鏡研究仙台病毒所引起的細胞融合

使用由子宮頸癌而來的HeLa糸田胞株做此實驗。此細胞表面利用掃描電子顯微鏡觀察時,表面構造不平坦,但無特別突起或構造。 利用紫外線非活化仙台病毒。此與細胞混合而置於37°C水浴中保持15分鐘後細胞表面變為平坦及細密。此細胞懸濁液若置於4°C,15分鐘後,細胞相互凝集且融合。再將此放入37°C水浴中振盪後細胞繼續進行融合。融合後產生的巨大細胞,表面由正在融合的細胞附著,但由於滲透壓之故表面構造漸變平坦及細密。

A HeLa cell line, which was established from a cervical cancer patient, was used for scanning electronmicroscopy. It showed an uneven surface but no remarkable projections or special structures were observed.The cells were fused by UV-inactivated Sendai viruse. When cells were incubated with the viruses in a 37°C waterbath, the surface of the cells showed a smooth and fine structure when compared with the control cells. Aggregation and also cell fusion were observed by keeping the for 15min at 4°C. Theseaggergated cells continued to fuse after shaking for 15 min in a 37°C waterbath. The fused giant cellsshowed an uneven surface at first and then the giant cells were observed with expanded surfaces.
