- 中文摘要: 本試驗之目的,乃利用放射性免疫測定法,探討 ACTH 對於雄鼠下視丘激性腺素釋放激素(GnRH)含量,直接或間接影響之機轉。割除腎上腺之雄鼠以0、10、20及40 I.U. ACTH分別處理0、4、8及16日,結果以不同劑量 ACTH 處理8日之各組,隨 ACTH 劑量之增加,對其下視丘 GnRH 含量之抑制愈明顯。其後正常或割除腎上腺之雄鼠,再以40 I.U. ACTH分別處理8日,結果均使其下視丘 GnRH 含量明顯降低。以上之結果顯示藥理劑量之ACTH會嚴重影響雄鼠之生殖,且可能同時涉及下視丘 GnRH 之合成或釋出。此外,ACTH對於下視丘 GnRH 之抑制作用,可能主要經由間接途徑,如腎上腺功能高亢,但外源性 ACTH 本身之直接作用,亦不容忽視。
- 英文摘要: The purpose of this study was conductedto investigate whether the effect of ACTH ondecreasing GnRH contents in the hypothalamusacts directly (by exogenous ACTH itself) orindirectly (mediated by adrenal glands). Toverify the effective dose and the duration oftreatment, the adrenalectomized male ratswere treated with 0 up to 40 I.U. of ACTHsubcutaneously daily for consecutive 0 to 16days, indicating that GnRH contents of thehypothalamus were significantly reduced bythe treatment with 40 1.U. of ACTH for 8days. Both intact and adrenalectomized malerats were treated with 40 I.U. of ACTH for8 days, the change of hypothalamic GnRHcontents, and the weights of testes and theaccessory sex glands were measured. Theresults showed that the hypothalamic GnRHcontents and the testes weights were reducedin both of intact and adrenalectomized animals,whereas the reduction of the weights ofseminal vesicle, ventral prostate gland andbody weight was merely inhibited in thentact animals but not in the adrenalectomized.These findings suggested that the reproductivefunction of male rats can be severely inhibitedby the pharmacological dose of ACTH andthe synthesis or even the release of GnRHof the hypothalamus may be involved. Mean-while, such inhibitory effect of ACTH onthe reduction of the hypothalamic GnRH maybe mainly mediated by the indirect effect, i.e. the hyper-function of adrenal gland butthe direct effect of exogenous ACTH itselfcan not be ignored too.
