激腎上腺皮質素對於大鼠排卵素抑制影響之研究

據許多文獻指出,動物長期使用ACTH處理後,其生殖機能有顯著之抑制作用,但此種抑制作用之機制猶未十分明瞭,本試驗之目的即在研討之。 以人工合成ACTH 0.2 mg每日一次注射於性成熟Long Evans品系之正常鼠及去腎上腺鼠,雄鼠連續注射14日,雌鼠連續注射14 ± 2日。雄鼠於最末一次注射之翌日上午9~11時,雌鼠則於動情前期斷頭犧牲之。秤量與體重及性腺之重,並分析其血中皮質固酮量、排卵素量及腦下垂體前葉排卵素量,據以評估ACTH之抑制性腺效應。 由試驗結果顯示ACTH對於正常雄鼠及去腎上腺雄鼠之生殖機能均有抑制作用,而對於雌鼠生殖機能之抑制作用則僅及於正常者。且此種抑制作用確已達腦下垂體,抑制其排卵素之合成與釋出。

Although many investigations have reportedthat the prolonged treatment of large dose ofexogenous ACTH inhibited the reproductivefunction of animals, the relationships betweenthem are still controversial. Therefore the pur-pose of this study was to settle the contro-versies. Cortrosyn Depot 0.2 mg (approximatelyequal to 20 I. U. of natural ACTH) was admini-stered daily to intact and adrenalectomizedsexually mature Long Evans Strain rats with 14consecutive days for male and 14 ± 2 consecutivedays for female. The male rats were sacrificedby decapitation between 0900 h and 1100 h onone day after the full, treatment. The sacrificetime of female rats was just on the proestrusstage. The results revealed that gonadal weights,serum LH concentrations and adenohypo-physial LH contents of intact and adrenalec-tomized male and the intact female were de-creased by the prolonged ACTH treatment.This inhibitory effects of ACTH on the repro-ductive system was evidenced to be involved inthe LH synthesis and release of pituitary gland.Furthermore the data showed that there aredifferent mechanisms in the inhibitory effects ofACTH on the reproductive system due to thesex differences.
