- 英文摘要: RanGTPase is importantly involved in diverse biologicalfunctions, such as nuclear transport, spindle formationduring mitosis, DNA replication and cell division.Thisreview summarizes yet another new role of Ran一controlof the activity of NF-kB in host immune responses. Over-expression of a specific allele of Ran results in preferen-tial accumulation of nuclear RanGTP, downmodulationof proinflammatroy cytokine production and protectionagainst septic shock. Nuclear RanGTP interacts with Ex-portin1,which in turn binds to IkB.Nuclear IkB binds toNF-kB in the nucleus, inhibiting NF-kB transcriptionalactivation of proinflammatory cytokine genes. NuclearIkB also functions as an adaptor between NF-kB andExportin1/RanGTP, enhancing export of NF-kB to thecytoplasm and further reducing the magnitude of hostimmune responses. From this picture, it appears thatmodulation of host immune responses via RanGTPasewill have an immense impact on a wide spectrum ofemerging infectious diseases, cancer and other geneticdiseases.
