- 中文摘要: 本研究目的在以 Tetracycline 抗生素浸漬,熱療處理以及電子顯微鏡觀察,究明柑桔黃龍病之病原菌質(Mycoplasma-like organism)之性狀。Achromycin(=tetracycline)100ppm 溶液浸漬罹病接穗15小時,對黃龍病病徵之發生俱有顯著的抑制效果,Aureomycin(=chlortetracycline)10 及100 ppm 溶液以及 Achromycin 10 ppm 溶液有輕微之治療效果,非 Tetracycline 抗生素之 Kanamycin 則無效,所有試驗抗生素對 Tristeza 病毒均無抑制效果。黃龍病複合病因之主要成員用 48℃以上到 54℃ 之熱水處理接穗10分鐘,會被破滅,但Tristeza 病毒成員不因熱處理而失去活性。由本省各種柑桔品種之病株而來之黃龍病病原分離株接種在椪柑或桶柑子實苗及嫁接在酸桔砧柑苗,在發病株之葉脈篩管中,常發現多數之多型態菌質體。在健康柑苗之葉脈篩管中則無菌質體存在。上述事實,顯示此菌質體可能為此病之主要病圍。菌質體之分佈不均,在老病葉之凸起葉脈中含量多。此個體之橫切片觀察,以球形或橢圓形居多,在縱切片中,有些篩管被密集之絲狀菌質體所佔滿。個體直徑在 90到 700 nm間,其中以300 nm為多,絲狀體之大小為90~400×200~2,000 nm。固體由三層單位膜包圍,其厚約15 nm(14~24 nm)。
- 英文摘要: Tetracycline test, heat treatment and electron microscopy were employed forclarifying the etiological nature of mycoplasma-like organism associated with citrusLikubin. Achromycin (=tetracycline) solution at 100 ppm completely inhibitedsymptom development of diseased scions dip-treated for 15 hrs. Aureomycin (=chlortetracycline) solution at 10 and 100 ppm and achromycin solution at 10 ppmshowed a little effect on suppression of disease development. The non-tetracyclineantibiotic, kanamycin, did not affect disease development. All antibiotics testedfailed to inactivate tristeza virus component. The main component of Likubinpathogen complex was inactivated by dipping the diseased scions in hot water attemperature higher than 48℃ up to 54℃ for 10 min. The heat-treatment did notinactivate tristeza virus component within scion sticks infected with the pathogencomplex. Numerous pleomorphic mycoplasma-like bodies were frequently detectedin vein sieve elements of Ponkan and Tankan seedlings or scion-Sunki stock com-binations artificially infected with several isolates of Likubin agent; no such bodieswere found in healthy seedlings. The distribution of the bodies was uneven in thesieve elements; more bodies were present in sieve tubes of old diseased leaveshaving swollen veins in comparison with those in younger diseased leaves. Fila-mentous bodies were occasionally found densely packed in sieve tubes in longitudinalsections. Spherical and ovoid forms of the pleomorphic bodies were present pre-dominantly in sieve cells of cross-sections. The body size was mostly 300 nm,ranging from 90 to 700 nm in diameter. The dimension of filamentous bodies wasmeasured'to be 40-90×200-2,000 nm. A trilaminar unit membrane with a thicknessof 15 nm(14-24 nm)encased the bodies.
