- 英文摘要: A number of infectious agents have been implicated inthe development of vascular diseases such as athero-sclerosis and posttransplantation arterial restenosis. Cy-tomegalovirus(CMV) has been reported to cause obliter-ation of coronary arteries by a progressive vasculopathythat involves proliferation of medial smooth muscle cells(SMC).In this study, we report that CMV enhances theserum-induced proliferation of human coronary SMCthrough activation of a superoxide-generating NADPHoxidase.Exposure of SMC to CMV for 2 h was associatedwith an 80%increase in NADPH oxidase. This increase inoxidase activity was associated with a two-fold increasein serum-induced DNA synthesis(5-bromo-2'-deoxyuri-dine incorporation)and significant interleukin-8(IL-8)production by SMC.Diphenylene iodonium,an inhibitorof NADPH oxidase, significantly inhibited CMV-inducedIL-8 production and promotion of serum-induced DNAsynthesis. Similar effects were seen following pretreat-ment of SMC with N-acetyl cysteine, a potent antioxi-dant, suggesting that oxidative stress following CMVexposure might be responsible for triggering the prolif-eration of SMC.From this study, we conclude that CMV-mediated promotion of SMC growth is redox sensitiveand may be mediated by NADPH oxidase.
