胎兒蛋白甲之純化及其抗體之製備

為了配合?免疫分析法 (Enzyme immunoassy) 的發展及高力價之特異抗體 (Highly specific
anti-α-fetoprotein antibody) 的製備,胎兒蛋白甲的純化工作為相當重要的一環。本報告簡單的
利用三種基本分離步驟:硫酸銨鹽析 (Ammonium sulfate precipitation),Con A-Sepharose 親力
色層分析 (Concanvalin A affinity-chromatography) 及製備電泳 (Preparative electrophoresis)
,由胎兒臍帶血 (Cord blood) 分離出高純度的胎兒蛋白甲,並以放射免疫分析法 (Radio im-
munoassay)、免疫電泳 (Immuno electrophoresis)、聚丙烯醯胺膠質電泳 (Polyacrylamide gel
electrophoresis) 來檢測每步驟之純度。並擬將所得之高純度胎兒蛋白甲皮下注射紐西蘭種白兔,
以製備高力價之特異抗體,進一步建立?免疫分析法 (Enzyme immunoassay)。

α-Fetoprotein has been isolated from cordserum by sequential use of ammonium sulfateprecipitation, concanavalin A affinity chromato-graphy and preparative gel electrophoresis. The product was checked by discontinuous poly-acrylamide gel electrophoresis, radioimmunoassayand immuelectrophoresis. The results showed that α-fetoprotein hasbeen enriched 100-fold and the major serumprotein albumin has been removed. Antiserumagainst the isolated product could precipitateauthentic α-fetoprotein. This antiserum can alsoimmunoprecipitate isolated product and cordserum but not normal human serum.
