本文報告新生大白鼠腦組織細胞之分離培養法及其結果。腦組織細胞在初代及繼代培養後的形態特徵可利用phase-
contrast microscopy, photomicrography和time-lapse-cinematography等方法加以觀察。
本研究之腦組織細胞培養之技術和體外長期培養之方式相當簡便,可獲良好之生長和增殖。
本培養法可使神經膠細胞群大量增殖並且亦可繼續在體外培養達30個月之久。
歷經一星期長時間的顯微電影法可以觀查到神經膠細胞之形態變化,可塑性以及明顯的動態。
在神經膠細胞突起上可觀察到?膜狀的膜構造,它經常波動或起伏運動,此可做為研判神經膠細胞突起和神經細
胞突起之重要依據。
神經膠細胞的胞體輪廓,突起及膜狀構造以Giemsa染色法可獲極佳之染色效果,值得推薦。
神經膠細胞與非神經膠細胞之間似有營養上之交互影響。
總之,本培養法適合繁殖大量的神經膠細胞並對探討神經膠細胞之形態和機能活動提供一種可應用之研究模式。
h - 英文摘要: Dissociated cell culture of brain tissue from neonatal rat was described. The morphology of braintissue cells in primary and passage cultures were examined by employing phase-contrast microscopy, pho-tomicrography and time-lapse cinematographic techniques. 1. The technique of preparing and maintaining these brain tissue cells in vitro was relatively easy andhad good condition of growth and proliferation. 2. This culture system was rich in neuroglial cells and was able to maintain in long-term culture upto 30 months in vitro. 3. A week long cinematography had revealed about the morphological transformation or plasticityof form and size and apparent motility of neuroglial cells. 4. Veil-like membranous expansion of neuroglial processes seemed to have ruffling or undulatingmovement and regarded as the evidence to distinguish neuroglial from the neuronal processes. 5. Giemsa stain was able to examine the definite silhouette of neuroglial cells especially on theirprocesses and membranous structures clearly. Hence it was strongly recommended. 6. Mutual trophic interaction between neuroglial and non-neuroglial cells was observed. This culture system was considered suitable to produce the neuroglial cell-enriched population andmight prove to be a valuable model for investigating neuroglial morphology and functional activity.
