國防醫學院生物化學系l - 英文摘要: It has already been established that eIF-3 has an ability to dissociate 80S ribosomes into 40S and60S ribosomal subunits and subsequently bind to 40S. However eIF-3 and 60S interactions have neverbeen demonstrated. In this study, Rabbit reticulocytes eIF-3 were coupled with Sepharose 4B for the purpose of creatingan affinity chromatographic column, and subsequently studying the interaction between eIF-3 and ribo-somes. It was then demonstrated that 60S ribosomal subunits binds to eIF-3. In comparing the 40S and60S ribosomes, this column preferentially binds 40S ribosomal subunits. The 40S ribosomal subunits werenot completely eluted until the KCl concentration was raised to 1.0 M. On the other hand, the 60S ribo-somal subunits could be easily eluted with 0.5 M KCl. When ribosomes that had never been exposed to0.5 M KCl (i.e., native ribosomes) were applied to the column, very little 40S ribosomal subunits were elut-ed with 1.0 M KCl. Furthermore, polysome derived ribosomes had similar properties to native ribosomes.These results suggest that 0.5 M KCl-washed ribosomes were different from native as well as from poly-some-derived ribosomes.
