- 英文摘要: Diabetes is a multifactorial disease that has now beenrecognized to involve overproduction of reactive oxygenspecies and pro-inflammatory cytokines. Peroxisomesare subcellular organelles with several important meta-bolic functions, and their role in the regulation of cellularoxidative stress is now well established.Despite havingtheir own antioxidant system,peroxisomes undergofunctional alterations during various conditions that areassociated with free radical production such as inflam-mation,ischemia-reperfusion,carcinogenesis and dia-betes. In this study we investigated the effect of diabeteson peroxisomal functions in rat kidneys and show for thefirst time that experimental diabetes induces redox-sen-sitive enhancement of peroxisomal activities. Streptozo-tocin-induced diabetes significantly increased(p<0.01)β-oxidation of lignoceric acid and the enzymic activity ofacyl coenzyme A oxidase.Catalase activity was signifi-cantly reduced(P<0.01)in the kidneys of diabetic rats,whereas the enzymic activity of DHAPATase《dihydroxy-acetone phosphate acyltransferase)was not markedlyaffected by diabetes. Treatment of diabetic rats withantioxidants, thiocetic acid and vitamin C attenuated thediabetes-induced modulation of peroxisomal functions.The present study shows that the diabetes-induced ef-fects on kidney peroxisomal functions are redox sensi-tive, and antioxidants might prove useful tools to alle-viate nephropathy in diabetes.
