- 英文摘要: Analysis of the genome of human herpesvirus 8(HHV-8)led to the discovery of several novel genes, uniqueamong the characterized gammaherpesviruses. Theseinclude cytokines(interleukin-6 and chemokine homolo-gues),two putative signal-transducing transmembraneproteins encoded by genes K1 and K15 at the genometermini,and an OX-2(CD200)receptor homologue thathad not previously been identified in a gammaherpesvi-rus. HHV-8 also specifies a diverged version of the gam-maherpesvirus-conserved G protein-coupled chemokinereceptor (vGCR)and a latently expressed protein uniqueto HHV-8 specified by open reading frame(ORF)K12.These cytokine and receptor homologues mediate signaltransduction or modulate the activities of other endoge-nous cytokines and receptors to enhance viral produc-tive replication,regulate latent-lytic switching,evadehost attack, or mediate cell survival.The viral signallingligands and receptors are also potential contributors tovirus-associated diseases, Kaposi's sarcoma,primary ef-fusion lymphoma,and multicentric Castleman's disease,and so represent potentially important targets for thera-peutic and antiviral drugs. Understanding these pro-teins' modes of action and functions in viral biology anddisease is therefore of considerable importance, and thesubject of this review.
