No intracellular HIV was seen in the wild-typecontrol, Density gradient analysis showed that the over-all density of these mutantvirions with excess PR (-)Gag-Pol was identical to that of the wild-type HIV-1. The find-ings indicate that overexpression of PR(-)GagPol, in thepresence of Gag synthesis, promotes intracellular bud-ding of the mutant virions and inhibits virus maturation·We have recently demonstrated that alteration of thehuman immunodeficiency virus type 1 (HIV-1) Gag/Gag-Pol ratio in virus-producing cells reduces the infectivityof progeny viruses and hinders the formation of stablevirion RNA dimers without impairing virion packaging ofthe viral genomic RNA. In addition,we have previouslyshown that the expression of GagPolmediates the se-lective packaging of tRNA . In this study we report thatoverexpression of uncleaved GagPol in the virus-produc-ing cell did not alter the packaging levels of tRNA .Similarly, altering the virion-associated Gag/GagPol ra-tio did not affect the virion packaging of the HIV-1 enve-lope protein nor cyclophilin A. Thin section electronmicroscopy analysis of the cells overexpressing pro-tease-defective [PR(-)] GagPol revealed immature viri-ons but no mature virions.
