- 中文摘要: The synthesis of a decaeoxyribonucleotide, d-CpCpApApTpTpTpTpGpG, as an exaple of DNA synthesis on a polystyrene polymer support using the phosphotriesters is described. The procedures for the recovery of the unreacted protected nucleoside phosphotriesters are also included. All operations and reactions were carried out in a 4ml polypropylene Bio-Rad Econo column fitted with 3-way teflon valve and a rubber septum. The average yield of each monomer addition is 96%. The decamer was obtained in high purity with an isolated yield of 56%. The purified decamer was characterized using a modified Maxam-Gilbert procedure.
