倉鼠連合下器官障蔽性質之超微結構研究

本報告係以靜脈內注射及腦室內射過氧?(Horseradish peroxidase, HRP)法與硝酸鑭溶液(Lanthanum nitrate solu-
tion)處理法,研究倉鼠連合下器官(Subcommissural organ, SCO)之障蔽性質(Barrier properties)。
經靜脈注射後之HRP,無法通過連合下器官之血管內皮細胞,因為該內皮細胞屬於非穿孔型(Non-fenestrated type)
,且具有緊密結合(Tight junctions)。但經由腦室內注射之HRP,可由腦室之表面進入連合下器官細胞間之細胞間隙及
腦室襯裹細胞(Normal ependymal cells)之細胞間隙;更可穿過此細胞間隙而達於其下方神經氈(Neuropils)部分之細
胞外間隙,終止於血管周圍間隙(Perivascular space),但未能通過內皮細胞。
以硝酸鑭溶液處理後,鑭金屬亦可進入連合下器官細胞之細胞間隙;但其進入之範圍?不如HRP之深,僅10-20
m。連合下器官附近,部分神經氈之細胞外間隙及血管周圍間隙,亦有鑭金屬之存在。
由以上之結果知:HRP與鑭追?劑不能通過連合下器官之血管內皮細胞,卻可通過連合下器官細胞之細胞間隙。
故吾人推論,倉鼠之連合下器官具有血一腦障蔽(Blood-brain barrier),但不具腦脊髓液一腦障蔽(Cerebrospinal fluid

Barrier properties of the hamster subcommissural organ (SCO) are studied by the intravenous and in-traventricular injection of the horseradish peroxidase (HRP) and by the method of lanthanum treatment.HRP, after intravenous injection, can not leave the capillaries lumen because of the interendothelial tightjunctions. After intraventricular injection, HRP enters the intercellular clefts from the ventricle throughthe intercellular junctions between the SCO cells and between the normal ependymal cells, and travelstoward the perivascular sheath, but never reaches the luminal surface of the endothelial cells. After the treatment of lanthanum nitrate solution, the lanthanum tracer penetrates into the in-tercellular junctions as the HRP does but to a lesser extent. The lanthanum tracer penetrates to a depthof 10 to 20 m from the ventricular surface. The extracellular space in the neuropils and the perivascularspace of the blood capillaries in the SCO area are also penetrated by the lanthanum tracer. The failure of the transport of the HRP and lanthanum tracer molecules to cross the endothelial cellsof the blood capillaries in the SCO and the penetration of the HRP and lanthanum tracer into the intercel-lular junction in the SCO area suggest the presence of the blood-brain barrier and the absence of cerebrospi-nal fluid (CSF)- brain barrier in the hamster SCO.
